Abstract

Acute pancreatitis is an inflammatory disease of the pancreas. It may leads to sever complication and high mortality without treatment. The pathogenesis is not fully understood, however the leukocyte activation, microcirculatory disturbances and oxidative stress are the major constituents of AP.
Various experimental models of acute pancreatitis have been developed in order to understand its pathophysiology, histological features, degree of severity and failure. it was shown that I.P. injection of high dose L-arginine induced pathological and histological changes consistent with acute pancreatitis. This non invasive model has been produced and explored extensively over the past decade, it shares features of disease progression seen in humans.
Alpha lipoic acid is a thiol antioxidant compound which demonstrated direct free radical scavenging properties. It can deliver antioxidant activity in both fat and water soluble mediums, and it is capable of having an antioxidant effect in both its oxidized (ALA) and reduced (DHLA) forms. It has a strong antioxidant with anti-inflammatory effects.
This study was carried out in order to investigate the possible effect of Alpha lipoic acid on L-arginine induced acute pancreatitis.
This study was carried on 4 main groups of adult male albino rats. The first of them is group I (control group) injected I.P. with 0.9% NaCl. Group II was injected with single I.P. injection of L-arginine in a dose of 250 mg/100 g body weight for induction of acute pancreatitis. Group III in injected with ALA in a dose of 1 mg/kg body weight/day for 7 days by I.P. injection then AP was induced by a single I.P. injection of L-arginine in a dose 250 mg/100 g body weight . Group IV, AP was induced by a single I.P. injection of L-arginine in a dose 250 mg/100 g body weight then after 24 h Rats were injected with ALA in a dose of 1 mg/kg body weight/day for 7 days by I.P. injection.
The parameters used to evaluate acute pancreatitis were serum amylase, lipase, CRP and histopathological changes of the pancreatic tissues.
The obtained results of this study could be summarized as follow:-
• L-arginine injection in a single dose of 250 mg/100g (I.P.); resulted in acute pancreatitis manifested by significant increase in serum amylase, lipase, CRP, histopathologic scoring and sever changes in the histopathological finding as compared with the control group.
• ALA injection in a dose of 1 mg/kg body weight/day for 7 days by I.P. injection then AP was induced by a single I.P. injection of L-arginine in a dose 250 mg/100 g body weight resulted in significant decrease in serum amylase, lipase, CRP, histopathologic scoring and also showed improvement of histopathological changes as compared with L-arginine injected group.
• When AP was induced by a single I.P. injection of L-arginine in a dose 250 mg/100 g body weight then after 24 h Rats were injected with ALA in a dose of 1 mg/kg body weight/day for 7 days by I.P. injection, there is significant decrease in amylase, lipase, CRP, histopathologic scoring with mild histopathological changes in pancreatic tissues as compared with that group injected L-arginine.
from the above results we conclude that I.P. injection of large dose of L-arginine produces acute pancreatitis and Alpha lipoic acid has a protective and beneficial effect againist L-arginine induced acute pancreatitis.
