Abstract

This study w~s designed to assess the protective effect of formalin-killed L.donovani promastigotes and EF of 1.don()vunt iri controlling the infection with the same organism in mice. I h.donovani promastigotes were grown in the 1°4N medium and the excreted factor was harvested from the overlay : df the culture. FCA was purchased commercially. ~ * Immunization was done SC, while the challenge infection was done through the IV route. After elapsing of a cal­ culated time animals were sacrificed and the parasite burden in liver and spleen was assessed. * The results obtained in this study s~ggested that the susceptibility of the mouse strain of the H.l.P.H. to 1.donovani infection (Sudanese strain) was lower than that of the highly susceptible strain (e.g.:BALB/C) of mice, but some-what higher than that of the less suscep­ tible or resistant strain. It was also suggested that the decrease (day 21 post infection) in parasite burden in this mouse strain was due to the acquired and not to innate immunity. * EF alone or combined with formalin-killed promastigotes initially enhanced the uptake of L.donovani promastigotes by liver and spleen of the mouse strain of the.H.I.P.H. * EF produced weak humoral ~nd cellular immu~e responses. * The results obtaineil with EF and P suggested that there might be apother immunological effector(s), other than the known cellular and humoral, in controlling leishmanial infection. if Results obtained with EF+FCA mixture were encouraging. ThiS””mixture modified the course of infection in J.iver and spleen in the mouse strain of the H.I.P.H. * The enlargement of organs (l~ver and spleen) in leishmania] infection seemed to be provoked by the immunological reaction rather than by the mere presence of the parasite in a particular organ. * P mixed with FCA yielded better results than when used alo~e for SC immunization against L.donovani infection. * In this study, the best results were obtained (a~ regards the parasite burden in liver and spleen) in the miLe immuuized with P+EF+FCA.
