Abstract
A Convenient One Pot Synthesis Of Pyrimido[4,Squinoline as S-Deaza Nonclassical Antifolates Inhibitors Refluxing a mixture of equimolecular amount of 6thioxotetrahydropyrimidine (1) benzaldehyde (2a) and dimedone (3) in absolute ethanol containing a catalytic amount of pipridine leads to the formation of pyrimidio[ 4,5-b ]quinoline 8a. The structure of 8a was established on the basis of its analytical and spectral data. The I H NMR spectra releaved a sharp singlet at b=4.75 ppm that integrated for one proton assigned to CH-5, in addition to signals for ring NH, aromatic, two methylene and two methyl groups (see Experimental). Moreover, 13C NMR gave strong evidence for compound 8a which showed signals forC-9a, C-I0a, C-4a and C-5a at 151.1,152.3,95.9 and 109.7 ppm, respectively. Similarly, compound 1 reacted with 2b-d and dimedone 3 to afford 8b.,d. In contrast to this behaviour, reaction of compound 1 with 2e and 3 led to the formation of 4e (Scheme 1).
