Abstract

Anti-epileptic drugs are a group of drugs used in treatment of different types of epilepsy. Extensive studies had been done about the teratogenic effect(s) of these drugs on cerebellum of different mammals including human.
Epanutin which is one of the most popular anti-epileptic drugs was found to cause many teratogenic effects on different organs including cerebellum.
Tegretol is one of the most famously used anti-epileptic drugs nowadays. But, however a more or less complete study was done on its effect(s) on development of cerebellum postnataly.
The study was a trial to detect the histological effects of the antiepileptic drug known as tegretol (caramazepine) on the histological structure of the albino rat cerebellum in the postnatal life.
This was done as a comparative study with the effects of epanutin(phenytoin). This work was designed to see any correlation between tegretol administration; compared with epanutin, to the pregnant females albino rat allover the period of pregnancy and changes occurring in the internal structure of the cerebellum concentrating on he cerebellar cortex of their offsprings.
This study was carried on 144 offsprings albino rats in the various stages of postnatal development up to the adult age (3 nonths).
Twenty four pregnant females albino rat were used and divided in o four main groups:
Group A (control group): Given nothing by mouth.
Group B (Sham control group): Given distilled water orally.
Group C (Experimental group I): Given epanutin orally in the maximal therapeutic dose allover the pregnancy period.
Group D (Experimental group II): Given tegretol in the maximal therapeutic dose allover the pregnancy period.
Offsprings dates used in the study were one, three, five, seven ten, fifteen, twenty one, thirty days old and three months old (adult).
Offsprings were anaesthesized with ether. Exteraction of brain from the skull of the offsprings at the used dates during the postnatal and adult period.
Preparations of serial paraffin sections for staining with the following stains.
1- Haematoxylin and Eosin.
2- Silver staining.
The control and Sham control groups showed the normal histological pictures of rats cerebellar cortex.
I- External granular layer A- Control group: The external granular layer was noticed to increase in thickness until it attained its maximal thickness around the date of 7 days old. Then gradually decreased in thickness till complete disappearance after the age of fifteen days postnatally.
B- Epanutin treated group: The external granular layer was thinner than that of control until the age of 7 days old. Then it attained its maximal thickness around the 10th day of age postnatally.
