Abstract

Schistosomiasis still represents a major health problem in many tropical and subtropical countries. Schistosomiasis mansoni occupies the first place in the list of endemic diseases in Egypt. Although the current treatment of schistosomiasis relies largely on praziquantel (PZQ), it has not significantly reduced the overall number of disease cases, perhaps due to inevitable resistance to praziquantel. In the present study, we evaluated the effect of various vaccination strategies in C57BL/6 mice including: 1- Single or multiple vaccination strategy with attenuated cercariae and subcurative dose (20 mg/kg) of PZQ. 2- Combination of single vaccination with subcurative dose of PZQ. 3- Passive transfer strategy of normal and vaccinated rabbit sera. 4- Comparing the reduction levels in number of recovered worms and ova count in liver and intestine. Also, the changes on recovered worms were studied by scanning and transmission electron microscopy. In addition, places of immunoglobulin binding of worms surface as an indicator for its intensity in immunized mice sera. Although RA generate high level of protection against subsequent challenge, this vaccine is not acceptable for use directly in human. The present study opted to use passive transfer of vaccinated rabbit sera (VRS) which induced reduction level of worms’ burden, ova count, and ultrastructural alterations in male and female worms. This level was very close to that induced by subcurative dose of PZQ. This may represent a hope in getting a promising strategy in schistosomiasis mansoni control.
