Abstract

Protease activity was investigated in cotyledons of 5-days old of marrow seedling. - Polyamines such as cadaverine, spermine, spermidine and putrescine stimulated protease activity. Treatment of cotyledons with different concentrations (0.2-1.0 mM) of HBL resulted in continous increase in protease activity. GA3 at a range of 100-200 µmol stimulated protease activity whereas as 100-300 µmol ABA reduced the induced-activity by GA3. Each of kinetin and BAP at 50-150 µmol induced protease activity in cotyledons. Jasmonic acid induced protease activity at a range of 25-150 µmol, but higher concentrations caused inhibition of the activity. The various algal extracts induced protease activity particularly Sargassum compared to Ulva and Corallina. - Both CdCl2 and PbCl2 inhibited protease activity and CdCl2 was stronger inhibitor. CdCl2 inhibited SOD, CAT and APX and increase MDA and ALA content decreased MDA content. The inhibition of protease activity was alleviated by HBL, JA, ALA and algal extracts. - Purification of protease was successful with specific activity of 200 U mg-1 protein. The optimum pH 8.0 and the optimum temperature of protease is 50 °C. - Carbohydrates such as sucrose, trehalose and raffinose increased thermostability of protease at 60 °C. Glycol chitosan and sarcosine increased thermostability of protease at 60 °C. NEM inhibited protease activity and the inhibition was reduced by casein as substrate. - Protease was immobilized on glass wool, Amberlite and chitosan. Chitosan was the best bead for enzyme immobilization. - Protease activity has detergent compatibility with Tide, Ariel and Persil. Protease activity succeeded to remove blood stain, digest cow meat, coagulate skimmed milk and hydrolyze blood clot.
