Abstract Forty female rabbits were randomized into four groups, ten animals each, group I (control), group II (Lidocaine-treated), group III (Irradiated) and group IV (Irradiated/lidocaine-pretreated). Both the animals of groups III and IV were irradiated with a single dose of 15 Gy but the animals of group IV had been received lidocaine pretreatment (Lidocaine hydrochloride (Xylocain, 2%, 1 mg/kg) before exposure to the dose of irradiation. Three, ten, thirty and sixty days the parotid glands were dissected and processed for H&E stain, immunohistochemical (ASMA) and transmission electron microscope. The irradiated group III exhibited cytoplasmic vacuolization, indistinct acinar outlines, inflammatory infiltration by H&E staining and massive loss of ASMA by immunohistochemical reaction. EM study of this group showed chromatin condensation, heterogeneous secretory granules, dilatation of RER and destruction of mitochondria. Indistinct changes were noticed in the lidocaine-treated irradiated group. The use of lidocaine, preserve the general cellular integrity and structural configuration of secretory apparatus during radiotherapy. |
