Abstract Seeds of Lupine, Chickpea and Pea were germinated in darkness for 6 days. Glycosidases were extracted from the obtained cotyledons with 0.2M sodium citrate buffer, pH 6.00, and then fractionated by ammonium sulphate to obtain the crude extract which contains the glycosidases enzymes. Enzymes β-galactosidase and α-mannosidase were found in Lupine extract, enzymes α-galactosidase, β-galactosidase, β-glucosidase and α-mannosidase were found in Chickpea extract, while enzymes α-galactosidase, β-galactosidase, α-glucosidase, β-glucosidase, α-mannosidase and β- mannosidase were found in Pea extract. pH optima for obtained enzymes was ranged between 3.00 - 6.00. Optimum substrate concentration was 50µM for the enzymes in Lupine extract, ranged from 25.00-50.00 µM for enzymes in chickpea extract, while it was ranged from 5.00- 50.00µM for enzymes in Pea extract. The Km value for β-galactosidase and α-mannosidase in Lupine extract was 5.50 and 0.90µM, while Vmax values were 4.80 and 3.20µM/min/ml. Km value for α-galactosidase, β-galactosidase, β-glucosidase and α-mannosidase in Chickpea extract was 7.50, 2.36, 3.70 and 2.00, while Vmax 2.20, 4.15, 2.20 and 2.90µM/min/ml, respectively. Km for α-galactosidase, β-galactosidase, α-glucosidase, β-glucosidase, α-mannosidase and β- mannosidase in Pea extract was 1.82, 3.70, 2.60, 2.07, 2.20 and 3.95µM, while Vmax values were 2.60, 6.90, 1.15, 1.55, 2.90 and 1.6 µM/min/ml. |
