Calcitonin gene-related peptide (CGRP), a major transmitter ill capsaicin-sensitive sensory nerves, is widely distributed in cardiovascular tissues and the release of CGRP is regulated by multiple autacoids including nitric oxide (NO). CGRP is broadly distributed in peripheral autonomic neurons innervating the cardiovascular structures including human heart. The physiological role of CGRP on cardiovascular system has not been fully established. There are several mechanisms by which CGRP produces vascular actions. Accordingly, this study was designed to explore the effects of CGRP on cardiovascular system both in vivo and in vitro and the possible mechanisms of action. I-In vivo studies: In this part of the study, a total number of 100, adult, male guinea pigs were involved. They were anesthetized by intraperitoneal injection of urethane; a polyethylene cannula was inserted in common carotid artery and was connected to pressure transducer through a three way valve for BP recording, the internal jugular vein was also cannulated for drug administration and the ECG limb cables were connected for ECG recording.
