Abstract Total of 35 New Zealand white (NZW) rabbit does with approximately 5.5 months of age and 3.25 kg live body weight (LBW) as well as 5 NZW bucks with average of 7.5 months of age and 3.75 kg LBW were used in this study. Rabbit does in the 1st group were i.m. injected with GnRH and slaughtered after 12 h from GnRH-injection (control group, G1). Does in G2, G3 and G4 were stimulated over-ovulation by i.m. injection with 75 IU of PMSG analogue (Foligon). Does in G2 were injected 48 h later by 150 IU of hCG, in G3 followed 48 h later by 0.02 ml GnRH analogue and in G4 followed 72 h by GnRH injection. All does in G2, G3 and G4 were slaughtered 12 h from the last injection for oocyte collection. Ovarian measurements and follicles more than 1 mm in diameter were recorded on each of left and right ovary of each doe. Oocytes were collected using slicing technique. Mature oviductal oocytes were collection from oviduct excised from superovulation does that slaughtered after 12 h following HCG or GnRH injection and the occurrence of ovulation. Oocytes were evaluated and classified into 4 categories. The recovery rate was determined as percentage of oocyte in proportional to each of the total vesicular follicles. Maturation in vitro rate of oocytes was evaluated using tissue culture medium (TCM-199) containing 10% (vol./vol.) from doe rabbit serum (DRS) or 0.3% (wt./vol.) from Bovine serum albumin (BSA) as compared to control medium (without serum) at 38.5oC for 20 hours. The current study could conclude that: 1) Using superovulation protocol of G3 (PMSG, GnRH 48 h later and slaughter after 12 h of GnRH treatment) resulted in the highest number of oocytes recovered from ovarian follicles and oviduct of rabbits (the highest recovery rates). 2) Adding 0.03 % bovine serum albumine to culture medium (TCM-199) resulted in higher maturation rate as compared to rabbit doe serum. However, the best quality of mature oocytes was obtained with (TCM-199) without serum. 3) Cleavage rate of in vivo maturation oocytes was higher than that of in vitro mature oocytes. |
