Abstract

The present investigation was carried out at the Tissue Culture Laboratory of Prof Dr. Helali, In vitro Tech. Lab., ADECO, Company, EI Mansoura, Egypt during the period or 1999 - 2002. The chemical analyses were conducted at the National Research Centre Labs. Dokki, Giza. Egypt. The experiments aimed to study some aspects of micro propagation of date palm (Phoenix daclylifera, L) Zaghloul cv. as affected by certain growth factors including type of explants, media and some growth substances. Direct organogensus by stimulating axillary buds formation from shoot tips of offshoot explants and indirect somatic embryogenesis by initiation callus from shoot apices, axillary buds and leaf primordia explants and germination of somatic embryos from ermbryonic callus were used (Plate 7). Browning and related phytochemical compounds as well as certain secondary products were examined. The most important results are summarized as follows: Direct organogenesis Effect of cytokinin 1. Shoot tip explants failed completely to form additional axillary bud during the first culturing periods up to 12 weeks. 2. Explants were able to produce axillary buds when cultured on MS media supplemented with either of BA or 2 iP especially at the 3rd culturing period. The interaction treatments showed an additive effect in this respect. Effects of auxins 3. Shoot tip explants failed completely to produced any additional axillary buds during the first 12 weeks over all auxins treatments. 4. At 18 weeks, the explants were able to produce axillary buds when cultured on MS medium supplemented with different concentrations of auxins (NAA, IBA). 5. There are an additive effect when different auxins used in combination.
