Abstract A total of 250 sample, 125 from each of apparently healthy Om EL-Khloul “Donax trunculus anatinus” and baclawese “Tartufo di mare” samples were randomly collected from Port-Said markets and examined for detection of some bacteria, sensitivity against some antibacterial discs and molecular identification and detection some of the virulent and toxigenic genes of the isolates. The prevalence of Aeromonas spp., Pseudomonas spp., Vibrio spp., E. coli and Staphylococcus aureus. from the examined Om EL-Khloul samples were 26/125 (20.80%), 35/125 (28.00%), 30/125 (24.00%), 0/125 (0.00%) and 20/125 (16.00%) respectively, while that of Baclawese were 36/125 (28.80%), 52/125 (41.60%), 40/125 (32.00%), 30/125 (24.00%) and 29/125 (23.20%) respectively. Biochemical and API20 identification of the isolates revealed that Aeromonas spp. were A. hydrophila, A. veronii, A. caviae and A. sobria, Pseudomonas spp., were P. aeruginosa, P. putida and P. florescent The identified Vibrio species were V. parahaemolyticus, V. vulanificus, V. anguillarum, V. alginolyticus and V. metschnikovil E. coli were not detected in Om EL-Khloul but detected in Baclawese. Staph. aureus was detected. Serological typing of P. aeruginosa were P. aeruginosa polyvalent III group E and P. aeruginosa polyvalent III group F. V. parahaemolyticus were identified as O3:K6, O4:K68, O4:K6, O1:K25 and O11:K36. E. coli was polyvalent I: (O26, O119 and O127) and Polyvalent II: (O125, O126). Staphylococcus aureus was identified serologically. Aeromonas showed Sensitivity to chloramphenicol and resist to all of the others examined antibacterial. Pseudomonas serovar were sensitive to most studied antibacterial except vancomycin, cefoperazone and ceftriaxone. Most of Vibrio serovar resist to studied antibacterial but sensitive to a lesser extent for amoxicillin, chloramphinicol and norfloxacin. E. coli was sensitive to some studied antibacterial agent and not sensitive for amoxicillin,gentamicin, erythromycin and cefatazidime, meanwhile staph. aureus was sensitive to studied antibacterial agent but resist to chloramphenicol and tetracycline. Molecular study showed that 16S rRNA were detected in Aeromonas. spp., Pseudomonas. spp., Vibrio. spp., and Staphylococcus aureus. spp., while E. coli strain was confirmed by phoA gene. Study of the virulence and toxigenic genes showed that Aeromonas species harbored Hemolysin and alt genes. toxA and oprL genes were detected in Pseudomonas strain, tdh and recA genes in Vibrio strain and sta and cfa/I genes were harbored in E. coli strain, but tst and clfA genes were detected in Staph. aureus. |
