Abstract

In the present study, genetic diversity among ten seeded (unknown) Date Palm genotypes growing at the Agricultural Research Center (ARC), Giza, Egypt was selected and used to investigate genetic diversity at the Agricultural Genetic Engineering Research Institute (AGERI) during 2021-2023, using SSRs, SCoTs, and DNA barcoding techniques. The current study was to provide more understanding of the genetic characteristics of these genotypes. The results from DNA fingerprinting revealed a broad sense of similarity among genotypes. All genotypes were sequenced and accessed into the NCBI GenBank database. The DNA barcoding revealed an efficient tool for studying phylogenetics in date palm genotypes. Overall, these approaches can be used not only to identify the seeded date palm genotypes but also to study the genetic diversity and phylogenetics of date palms. The best approach was DNA barcoding via rbcl gene. It can be said that this gene is a universal barcode for date palm genotypes.
