Abstract

The aim of the research is to study the effect of cisplatin injection on the
histological structure of both liver and kidney of the adult albino rat and also the possible protective role of melatonin.
The results of the current study revealed that:
 In hematoxylin and eosin stained sections: cisplatin administration caused marked changes in the renal tissue like, glomerular congestion, atrophy and distortion, interstitial hemorrhage. Tubular distortion, cellular
vacuolation, epithelial flattening, intraluminal cellular and hyaline casts
and inflammatory mononuclear cellular infiltration. There were also
histopathological changes in the liver of the rats. These changes were
represented by congestion of the central veins and the blood sinusoids and also congestion of the portal veins. The hepatocytes were also exposed to degenerative changes in the form of the appearance of vacuoles in the cytoplasm; pyknotic nuclei and apoptosis of some cells. There was also
inflammatory cell infiltration. Co-administration of melatonin with
cisplatin resulted in marked improvement of the previous morphological changes.
 In mallorys trichrome stained sections: Cisplatin increased deposition of collagen fibers as compared to the control and melatonin groups in both liver and kidney sections. Co-administration of melatonin with cisplatin
resulted in marked reduction of collagen fiber deposition to become
relatively similar to control.
 In PAS stained sections: Administration of cisplatin resulted in a weak PAS reaction in hepatocytes reflecting depletion of glycogen stores. Also there is a weak reaction in the kidney sections demonstrated by partial or complete loss of apical brush border. The glomerular capillaries’ and renal tubules’ basement membranes were also shown to be disrupted. Coadministration of melatonin with cisplatin resulted in marked improvement in the form of preserved reaction in both liver and kidney sections.
 Immunohistochemical study of activated caspase-3 and iNOS in both liver and kidney sections showed a negative immune reactivity in the control and melatonin groups, significant increased immune reactivity in the cisplatin-group compared to the control group while co administration of
melatonin with cisplatin significantly decreased immune reactivity as
compared to the cisplatin group.
