Abstract

Comparing the effect of transplantation of mesenchymal stem cells (MSCs) against adult hepatocytes in an experimental model of chronic liver failure to reach the best cell - based line of treatment for chronic liver diseases. MSCs that were isolated from human cord blood (CB) were cultured, propagated and characterized. Both human CB - MSCs and freshly isolated rat hepatocytes were transplanted into a CCL4 cirrhotic experimental model. Animals were sacrificed 3 month after transplantation, and liver functions were assessed. Liver specimens were subjected to H and E, Masson{u2019}s and Sirius red staining for histopathologic analysis as well as immunohistochemistry staining for albumin, AFP and prox1. Compared to the cirrhotic control group (gp); both hepatocytes gp and MSCs gp showed significant improvement of ALT, AST, Albumin and bilirubin levels. Hepatocytes engraftment resulted in a higher albumin level while MSCs engraftment showed better improvement in ALT, AST and bilirubin levels. Compared to the cirrhotic control group (gp); histopathologic examination of both MSCs gp and hepatocytes gp showed resolution of the fibrous septa surrounding cirrhotic nodules and improvement of the hydropic degeneration. Successfully engrafted cells were confirmed by the brownish cytoplasmic staining of Prox1 in rat hepatocytes and AFP and Albumin in human MSCs
