Abstract Granisetron hydrochloride proniosomes were prepared by slurry method using maltodextrin as a carrier. The prepared proniosomes were incorporated into buccoadhesive tablets employing mucoadhesive polymer (xanthan gum, HPMC or carbopol® 974P). In-vivo pharmacokinetic study revealed higher bioavailability of the selected buccal formulation relative to conventional oral formulation of granisetron. In situ nasal gels combining granisetron hydrochloride and ketorolac tromethamine were prepared. The analgesic effect of the selected formulation was assessed by two different nociceptive tests: Formalin (chemical) and hot plate (thermal) tests. The results of pharmacological study showed that intranasal administration of the selected formulation produced a significant analgesic effect against chemical and thermal nociception as compared to intra-peritoneal one |
