Abstract Several studies have shown associations of ADIPOQ polymorphisms with diabetes mellitus, obesity and cardiovascular diseases; however, inconsistencies are present with different study populations. The aim of the present study was to indicate the relation between adiponectin gene mutation variant +276G/T (rs1501299) and risk of T2DM among Egyptian obese patients. This case-control study included totally 100 Case , they were subdivided into four groups ;group 1 included 25 nondiabetic non obese cases ,group 2 included 25 diabetic obese cases ,group 3 included 25 nondiabetic obese cases and group 4 included 25 diabetic non obese cases . They were recruited and assessed for eligibility from the Endocrinology Outpatient Clinics and Inpatient Ward in Menoufia University Hospital. The present study revealed a statistically significant increase in weight and BMI among the diabetic obese group B and the non diabetic obese group C in comparison with the diabetic non obese group D (P value=0.001). Moreover, there was a statistically significant increase in fasting blood glucose, 2h postprandial blood glucose and HbA1C % among diabetic obese group B and the diabetic non obese group D in comparison with the non diabetic obese group C (P value=0.001). The present study revealed statistically significant difference between the studied groups regarding TSH levels (P value>0.001). Regarding the liver function test among studied groups, the present study indicated a statistically significant increase in ALT among nondiabetic obese group C in comparison with the diabetic obese and the diabetic non obese groups (P value=0.001). Summary 63 The present study indicated statistically significant difference between the studied groups regarding blood urea and creatinine levels (P value 0,001). The present study indicated a statistically significant increase in serum cholesterol, HDL, LDL and serum triglycerides among diabetic obese group B and the non-diabetic obese group C in comparison with the non-diabetic non obese group A and the diabetic non obese group D (P value=0.001). The present study indicated statistically significant difference between ADIPOQ SNP by real time PCR among studied groups. (P value 0,003). The present study revealed that the frequency of TT genotype for rs1501299polymorphism was significantly associated with increased height (P value=0.013), fasting blood glucose (P value=0.002), 2h postprandial blood glucose (P value=0.024), TSH (P value=0.001) and creatinine (P value=0.026) among patients in the diabetic obese group B. The present results revealed that the frequency of TT genotype for rs1501299polymorphism was significantly associated with increased weight (P value=0.005), BMI (P value=0.001) and serum triglycerides (P value=0.008) among patients in the non-diabetic obese group C. The present study revealed that the frequency of TT genotype for rs1501299polymorphism was significantly associated with increased fasting blood glucose (P value=0.004) among patients in the diabetic non obese group D. |
