Oral route is the most preferred route for systemic delivery of drugs, although, it has a lot of disadvantages as, degradation of sensitive drugs in gastrointestinal tract, gastric irritation, poor absorption of drugs and hepatic first pass metabolism. Buccal bioadhesive preparations are a good alternative for oral route, avoiding all of its drawbacks. Liquisolid compacts is one of several methods used for the enhancement of drug solubility, where a liquid may be transformed into free flowing readily compressible and apparently dry looking powder by simple physical blending with selected carrier and coating materials . Famotidine is an H2-receptor blocker, very slightly soluble in water, has a short elimination half life time of 3 hrs, with low absolute bioavailability of about 40 to 45%, which may be attributed to minimal first- pass metabolism and its susceptibility to gastric degradation. The aim of this work is the formulation of famotidine as buccoadhesive cup and core tablet, characterized by rapid release of the cup loading dose formulated in a more soluble form, followed by an extended release of the core maintenance dose. Also, increasing the bioavailability by escaping first pass effect, and avoiding gastric degradation.
