Abstract

This study aimed to explore if the monosialoganglioside GM1 could mediate neuroprotection in global ischemia/reperfusion (I/R) insult and to investigate the role of tropomyosin related kinase (Trk) receptor, p75 neurotrophin receptor (p75NTR), and the pro and mature forms of nerve growth factor (NGF) in mediating GM1 effects. This study also tested if GM1 effects could be modulated by concurrent administration of the flavanol Epicatechin (EPI). Global ischemia was mediated by occluding both carotid arteries for 30 min., which was followed by a one week reperfusion period. Measured effectors of Trk and p75NTR increased with the used treatments, and the burst of pro/mature NGF ratio that was observed a week after I/R was effectively reduced by all treatment regimens. This was associated with an improvement in both memory retention and acquisition functions, as observed by the Morris water maze test
