Abstract

Moringa oleifera is one of miracle tree and great interested medicinal plant which belongs to Moringaceae. It is widely used as a nutritive herb and possesses valuable pharmacological activities. In this study, a rapid, sensitive and accurate HPLC method for qualitative and quantitative determination of ascorbic acid, gallic acid and rutin in two different solvent, methanol and ethanol, extracts of Moringa oleifera leaves cultivated in Egypt was investigated. The separation was performed and validated by both gradient and isocratic method utilizing C18 column with a mobile phase 10% (v/v) acetonitrile and 1.0% aqueous acetic acid mixture for determination of ascorbic and gallic acids, and 60% (v/v) methanol and 1.0% aqueous acetic acid mixture for determination of rutin. Moreover, a successfully spectrophotometric quantification of total phenolics and flavonoids content in the plant extract was achieved. The higher concentrations of ascorbic acid 53.20 μg/mL was observed in 70% ethanolic extract, while the higher concentrations of gallic acid, 51.20 and 30.26 μg/mL, were observed in the 50% methanolic and ethanolic extracts; also the higher concentrations of rutin, 60.00 and 59.93 μg/mL, were observed in the 100% methanolic and ethanolic extracts. The validation of the method was performed where it demonstrated acceptable precision with relative standard deviation (RSD%) values lower than 3.0%.
