Abstract

Simeprevir sodium (SMV); a novel hepatitis C inhibitor, represses hepatitis C viral replication by binding to and inhibiting the protease, hepatitis C virus (HCV) NS3/4A. In this way, it is known as a prompt acting antiviral agent. Two simple and highly sensitive spectrophotometric methods have been developed for the quantification of simeprevir sodium (SMV), in the pure form and in pharmaceutical formulations, as a single component utilizing direct, standard addition, and ion-pair formation methods and in a mixture with other antiviral drugs using H-point standard addition method (HPSAM). HPSAM showed suitable abilities to resolve accurately overlapped absorption spectra of the compounds by simultaneous determination of SMV in its binary and ternary mixtures with Sofosbuvir and/or Ledipasvir. Ion-pair formation methods which involves the reaction of SMV with some sulphonphthalein dyes, e.g. bromothymol blue (BTB), bromocresol purple (BCP), bromocresol green (BCG), and bromoxylenol blue (BXB). The reaction conditions were studied and optimized. The proposed methods are rapid, accurate, sensitive, selective and cost effective. They were successfully applied to the analysis of commercial tablets as Olysio® and Sofolanork plus®. Statistical treatment of the results was performed with regard to accuracy and precision using student{u2019}s t- and Ftests at 95% confidence level
