Abstract

Poly (ADP-ribose) polymerase (PARP) is a keyenzyme that regulates the cell survival in many pathological conditions. we investigated the effect of 3-aminobenzamide (3AB), a PARP inhibitor, in different doses to achieve the benefit of PARP inhibition on multiple organ damage induced by exposure to ionizing radiation (IR) without losing the beneficial role of PARP enzyme in repairing the DNA breaks. Rats were treated intraperitoneally with (3AB) in a dose of 5,10 and 15 mg/ kg, 1 hour prior to irradiation at a level of 6 Gray (Gy). Control groups were run concurrently. Rats were sacrificed 24 hours after irradiation. IR led to changes in the level of oxidative stress biomarkers (elevation of thiobarbituric acid reactive substance (TBARS) and nitrite associated with a decrease in total antioxidant capacity as well as in the level of inflammatory parameters (elevation of myeloperoxidase (MPO) associated with expression of cyclooxygenase-2 (COX-2). Irradiation caused cellular damage characterized by an increase in serum lactate dehydrogenase (LDH) activity and cytosolyic Ca+2, apoptotic changes characterized by expression of Caspase-3 as well as decline in complex-I activity and adenosine triphosphate (ATP) level. Pre-treatment with the small doses of 3AB guarded against the changes in all the measured parameters, conversely pretreatment with the dose of 15 mg/kg showed no effect on the damage induced by irradiation in the selected tissues.Moreover, 3AB has a dose dependent effect on viability of Vero cells. Low doses of 3AB exerted a protective effect againstradiation induced multiple organ damage
