Immunohistochemical study on the effect of adipose-derived stem cells versus clomiphen on treatment of experimental polycystic ovary in rats

Abstract
Background and aim of work: Polycystic ovarian syndrome (PCO) is a syndrome of ovarian dysfunction along with the cardinal features of hyperandrogenism and polycystic ovary morphology. Clomiphen is the first line of treatment, however, it is associated with many side effects.Therefore, this work aimed to compare the effect of adipose-derived stem cells (ADSC) to clomiphen on the treatment of experimentally-induced polycystic ovary in rats. Materials and methods: 50 adult female albino rats were divided into: group I (control), group ЇЇ (PCO group) that received Letrozole for 21 days. Then 5 rats from each group were sacrificed to prove PCO. Remaining 20 rats of PCO group were subdivided: Subgroup ЇЇB (Recovery group), Subgroup ЇЇC (clomiphene-treated group), Subgroup ЇЇD (ADSC-treated group), Subgroup ЇЇE: (clomiphene + ADSC treated group), while the control group was subdivided into subgroups IB, IC, ID and IE, that received the solvents for 5 weeks. Ovarian specimens were processed and stained with H & E, Alcian blue stain, Periodic acid Schiff (PAS) reaction, immunohistochemichal stains using Ki 67 and caspase 3 antibodies. Blood tests, morphometric and statistical analyses were done. Results: Clomiphen and ADSC improved hormonal profile, ovarian and body weight and improved also the ovarian histological architecture where various stages of ovarian follicles were noticed, including corpora lutea and few cysts. Conclusion: Clomiphen effect was found to be significantly better than ADSC, while combining ADSC + Clomiphen gave the best results
