Abstract

The present study was designed to investigate the acrylamide (ACR) content in some Egyptian food, (falafel, fried Potatoes, fried eggplants and fried onions) as well as the mitigative effect of quercetin (QE), rutin and ferulic acid compounds on ACR content. Acrylamide content in final product was quantified by HPLC. The effect of quercetin compound on ACR toxicity was also evaluated in experimental animals. The rats were feeding on basal diet and received oral 20 mg/kg b.w of ACR with 100, 50 and 25 mg/kg w.t of quercetin; positive control group (ACR group) received 20 mg/kg b.w of ACR for 30 days. The results showed that ACR was found in falafel, fried potatoes, fried eggplants and fried onions. The quercetin had the highest mitigative effect on ACR content in homemade potatoes crisps at (Hb%) concentration, red blood cell (RBC) count, hematocrit (HCT), total protein (T.P), albumin (ALb), acetyl cholinesterase (AChE), testosterone total, testosterone free in serum were decreased in rats-treated with ACR. While alanine aminotransferase (AST), aspartate aminotransferase (ALT), urea, creatinine, lipid peroxidation (LPO), 8-hydroxydeoxy guanidine (8-OHdG), protein carbonyl (PC), reactive oxygen species (ROS), glutathione peroxidase (GPx), tumor necrosis factor- (TNF-α), nitric oxide (NO), myeloperoxidase (MPO), caspase-3, Bax, P53 and cytochrome-C in liver, kidney, brain and testes tissues were increased in rats-treated with acrylamide induced inflammation and oxidative stress. The harmful effect of ACR was attenuated by quercetin. The indicate histopathological changes in liver, kidney, brain and testes were elevated after treatment with ACR. However, quercetin ameliorated acrylamide histopathological alternations. These results indicated that quercetin (QE) had a promising protective effect on ACR content in food stuffs as a result of its mitigative action on acrylamide in homemade potatoes crisps. In addition it had a protective effect against ACR induced inflammation and oxidative stress in rats.
