Abstract

This study was carried out mainly in the Department of Agricultural Microbiology, Fac. Agric. Zagazig Univ., and Central Lab. for Aqua. Res., Abbassa, Abu-Hammad, El-Sharkia, Egypt, during 2018-2020 in order to study the prevalence of Pseudomonas spp. in agricultural wastewater as well as in soil in El-Sharkia Governorate. Also, this study concentrated on the counting of different Pseudomonas spp. to take an overview about the bacterial load of some agricultural wastewater as well as in soil samples. In addition, four Pseudomonas aeruginosa (ATCC 27853 THL) phages were isolated from different agricultural wastewater and soil samples and characterized with electron microscope. Some factors affecting phages were performed also. Molecular characterizations and in vitro phage cocktail application as well as phages (Ps1 and Ps2) therapy against pathogenic Pseudomonas aeruginosa (ATCC 27853 THL) were also studied and the results could be summarized as follows:
1- The total bacterial counts and Pseudomonas counts were examined in soil. The total counts of bacteria ranged from 3×106 to 4.12×109 CFU g-1dw.soil, Pseudomonas count ranged from 2×102 to 5.2×103 CFU/ml in water samples. However, the highest average count was found in soil sample collected from Mashtoul El-Souk (Vicia faba) (4.12×109 CFU g-1dw.soil), while the lowest average count was found in Oryza sativa soil (Abo-Kebeer).
2- Eight selected isolates were identified at Academic Park Fac. Medicine, Alex. Univ., Egypt, using MALDI-TOF-MS namely Pseudomonas aeruginosa 19955_1CHB, Pseudomonas fluorescens DSM50090T HAM, Pseudomonas corrugate DSM7228T HAM, Pseudomonas jinjuensis LMG21316T HAM, 2 Pseudomonas aeruginosa ATCC 27853 THL, 2 Pseudomonas caricapapayae LMG2152T HAM.
3- Using 16S rRNA sequence for the confirmation of an identification of Pseudomonas aeruginosa (ATCC 27853 THL), was done in Animal Health Research Institute, Giza, Egypt. This isolate was conformed as Pseudomonas aeruginosa strain K11.4 16S ribosomal RNA gene.
4- Pseudomonas aeruginosa ATCC 27853 THL, Pseudomonas aeruginosa 19955_1CHB and Pseudomonas fluorescens DSM50090T HAM were highly sensitive to antibiotics (aztreonam, cefoxitin, levofloxacin and streptomycin).
5- Pseudomonas aeruginosa ATCC 27853 THL as well as Pseudomonas aeruginosa 19955_1CHB recorded the highest MAR indices giving 0.61.
6- Many phages were isolated from different sources of agricultural wastewater and soil using Pseudomonas aeruginosa (ATCC 27853 THL) as host. Four phages were chosen namely Ps1, Ps2, Ps3 and Ps4.
7- Among the six bacterial isolates tested the lytic phages (Ps1, Ps2, Ps3 and Ps4) were infectious to four or three different bacterial strains.
8- None of the four phages could lyse another bacterial genus tested including Aeromonas veronii (B565), Aeromonas hydrophila (DSM30187) Aeromonas caviae (CECT4221), Aeromonas encheleia (CECT4342) and Aeromonas salmonicida
