Abstract

Domperidone is a dopamine D2 receptor antagonist. It has a low oral bioavailability of about 13{u2013}17% because of the low water solubility. It also has a short biological half-life. The short half-life favors the idea of developing a sustained release dosage form. On the other hand, domperidone is a weakly basic drug that may encounter release challenges from the conventional sustained release formulation when exposed to the increasing pH of the intestinal fluid . The drug has better solubility in the acidic pH of the gastric fluid . Hence, the formulation of domperidone in a gastro-retentive drug delivery system could be a potential solution to overcome the problems of low oral bioavailability, short biological half-life, and poor solubility in high pH. The gastro-retentive technique depends mainly on two approaches, to allow the formulation to remain in the stomach for a prolonged time at the low pH, and to allow the slow release of the drug from the formulation. In the present work, the main objective is to develop and characterize a domperidone gastro-retentive delivery system to enhance its oral bioavailability, control the plasma drug level, decrease the dosing frequency, and improve patient compliance
