Abstract

Thesis is focused on the determination of mepivacaine hydrochloride (MPVHC) as a local anesthetic drug individual or with combination with levonordefrin (LVN). A potentiometric method based on modified carbon paste ion-selective electrode and chromatographic methods using UPLC and LC/Ms/Ms instruments were described for the determination of MPVHC in different pharmaceutical and pure samples.The potentiometric method using carbon paste (MCPE), the influences of the paste composition, different conditioning parameters and foreign ions on the electrode performance was investigated and response time of the electrode has been studied. The electrode showed Nernstian response of 57.72 mV decade-1 in the concentration range of 3.1{u00D7}10-7{u2013}1{u00D7}10-1 mol L-1 for MCPE.The electrode was found to be usable within the pH range of 3.0{u2013}8.0, exhibited a fast response time (about 14s), low detection limit (3.1{u00D7}10-7 mol L-1), long lifetime (110 days) and good stability for MCPE (Electrode I).The electrode was successfully applied for the determination of MPVHC in pure solutions, pharmaceutical preparation, urine and plasma samples. The results obtained applying this potentiometric electrode is comparable with British Pharmacopeia.The method validation parameters were optimized and the method can be applied for routine analysis of MPVHC drug
