Abstract

The objective of this work was to indicate the possible synergistic effect of this combination on the cancer stem cell population using an in vitro model of metastatic breast cancer cell line MDA-MB231. This aim was carried out by investigating the crosstalk between two of the cell death pathways of cell autophagy and apoptosis and their effects on stemness of breast cancer. the study was designated to be divided into six main groups. The effect of BSG on cell proliferation and assessment of its IC50 was tested using MTT assay. BSG showed a significant inhibition at cell growth and survival of MDA-MB231 colonogenic assay. In the current study, BSG has not only induced cell apoptosis but also induced autophagic cell death which is dependable not only on the BSG dosage but also on time of exposure. In conclusion, apparently, the cells exposed to IC50 dosage were under stress that exceeded the threshold of stress that the cells can withstand; so, cell apoptosis was induced and the autophagic pathway was blocked leading to a downregulation at cancer cell invasiveness and stemness.
