The Protective Effect of Taurine on Oxidative Damage Induced by Ipomea Carnea in Wistar Male Rats.

Abstract

The adverse deleterious influences of Ipomea carnea on brain tissue and relative ability of Taurine to prevent this induced neurotoxicity has been examined. Seventy five Wistar male rats weighing 175±2 g were used. The rats were allotted into five groups (n = 15 per each) as follows: Control group: that received saline I.P. injection daily for eight weeks. Ipomea carnea extract (ICE) group, in which rats were orally supplemented for two months with ICE (15 mg/kg b.w /day). Taurine treated group, in which rats were administered for “two months” Taurine (200 mg•kg−1, I.P.) daily. Taurine + ICE group, in which rats were supplemented with (200 mg•kg−1, I.P) for one month, then ICE (15 mg/kg b.w /day) for one month. ICE + Taurine group, in which rats were supplemented with ICE (15 mg/kg b.w. /day) for one month and then (200 mg•kg−1, I.P) for four another one month. The results showed that Ipomea carnea extract increased Intracellular ROS levels. However, Taurine supplementation significantly reduced ROS levels in injured brain cells. And Elevation in lipid peroxidation marker (MDA) and perturbation in the antioxidant homeostasis in addition to increase the levels of brain and serum cholinesterase (ChE), Alpha total creatine phosphokinase (CPK), also creatine phosphokinase isoenzymes BB (CPK-BB) and lactate dehydrogenase (LDH). With down-regulation of reduced superoxide dismutase (SOD), catalase (CAT), glutathione (GSH) and activities and gene expression. While Taurine supplementation significantly decreases lipid peroxidation in injured brain cells. Furthermore, increasing GSH content, SOD activity, CAT activity, in injured brain cells. Alpha total creatine phosphokinase (CPK), Isoenzymes creatine phosphokinase BB (CPK-BB) and also lactate dehydrogenase (LDH) and these all of effects were ameliorated by Taurine. ICE induced up-regulation of TNF-α (inflammatory cytokine) while following the treatment of Taurine (TNF- α) levels were decreased. ICE substantially cause down-regulation of nuclear factor (erythroid-derived 2)-like 2 (Nrf2) that is a very crucial component that regulates the antioxidant enzyme transcription in the ICE - challenged rats group, and all of these effects were ameliorated by Taurine. In addition to Histopathology investigations (H&E), Transmission Electron Microscope (TEM), and Immunohistochemical brain examination (GFAP) protein, these findings support our findings that proved the protective role of Taurine as a supplement could recover Ipomea carnea -induced oxidative changes and brain damage. Reproductive abnormalities appear to be potentially important in poisoning with Ipomea carnea. The cytological observations revealed that the extracts of ICE had a strong mitodepressive and different stages of cell division along with the chromosomal irregularities. The low quality of the semen poisoned represented in sperm abnormalities by ICE. Supplementation of Taurine could recover Ipomea carnea Extract induced oxidative stress changes and brain damage. And reproductive degeneration, sperm and chromosomes preservation.
