Abstract

The present study aimed to study genetic polymorphisms in folate-metabolizing gene MTR (methionine synthase) (A2756G) and V600E in BRAF gene in a sample of Egyptian women to investigate their roles in breast cancer susceptibity within the sample size. The study included 200 women (100 breast cancer female patients and 100 healthy females as a control). The breast cancer female patients were obtained from the Medical Research Institute, Alexandria University, Egypt. The two selected polymorphisms were MTR A2756G (rsl805087) and BRAF V600E (rsl 13488022). Genomic DNA was isolated from whole blood for genotyping the two selected polymorphisms among the participants. PCR-RFLP was carried out to genotype MTR A2756G using a Haelll restriction enzyme. While, BRAF V600E was genotyped by ARMS method. Total RNA was extracted to determine the gene expression of MTR A2756G and BRAF V600E by using qRT-PCR technique among this study. In the present study, there was no significant correlation between the breast cancer and other clinicopathological parameters (e.g., family history, smoking, breast feeding, age, and histological grade). There was no significant difference in the frequency of the AG genotype of MTR A2756G between breast cancer patients and control subjects. Additionally, there was no significant difference in the frequency of A and G alleles of MTR A2756G between the breast cancer patients and the control subjects. Also there was no significant difference in the frequency of the AT genotype of BRAF V600E between breast cancer patients and control subjects. There was no significant difference in the frequency of A and T alleles of BRAF V600E between the breast cancer patients and the control subjects. With regard to the expression level of MTR, data revealed that there was no significant difference in its expression between tumor tissues and adjacent normal tissues of breast cancer patients. A statisctical correlation in MTR expression levels between tumor tissues and normal adjacent tisssues of breast cancer patients was not evidenced. On the contrary, a statisctical correlation in BRAF expression levels between tumor tissues and normal adjacent tisssues of breast cancer patients was evidenced.
