Abstract

Cloning and Expression of Hepatitis C Virus (HCV) Nonstructural 3 (NS3) Region of Genotype 4a to Study the Clearance Versus Persistence of Viral Infection.

Serologic tests are essential for the management of hepatitis C virus (HCV) infection in order to diagnose infection, select treatment and assess virologic response. In order to help in construction an epitope map for type 4a HCV to aid in the development of improved immunodiagnostic reagents, we have selected full length non-structural region 3 (NS3) peptide of the HCV polyprotein, about 631 amino acids for expression and evaluation of immunodiagnostic potential. The selected peptide was expressed, and its immunoreactivity towards the sera from patients infected with HCV-4a was assessed using Western blot and ELISA. The reactivity of the expressed peptides was compared with a commercially available ELISA kit. The peptide antigen was well recognized by HCV infected sera, by western blot. Furthermore, the NS3 peptide was sensitive in detecting the antibody in patient sera as the commercial ELISA kit this result is useful as we used only one peptide although in the commercial kit (Abbott HCV EIA version 3), the recombinant used are (HCr43, c-200, c-100-3, NS5) corresponding to core-NS3, NS4, NS5 regions of HCV polypeptide. The results demonstrate the presence of epitopes on the selected peptide which is potentially useful for the development of improved immunodiagnostic reagents for HCV type 4 and to demonstrate studies for vaccine development.
