Abstract

The current study was conducted to evaluate the protective role of ALA on aging in rats and changes occur.This study has been applied on Forty eight wistar albino male rats.Twenty four of them were young about 2weeks old and the other twenty four were adult about 4 weeks old. The rats were divided into four equal groups as follow: group I: (control group) Include twelve young male rats fed on normal diet GroupII:(control group This group has twelve adult rats fed on normal diet GroupIII: (lipoic acid group Comprised of twelve young male rats fed on lipoic acid rich diet days.group IV: (lipoic acid group Include twelve adult rats fed on lipoic acid rich diet. At the middle and at the end of experimental period (2weeks) half of rats were fasted overnight , anesthetized with diethyl ether and blood samples withdrawal through cardiac puncture from the heart, immediately the blood sample were divided into sterile, dry, capped tubes. serum separation. The tubes was left in vertical position at room temperature then centrifugation at 3000 rpm for 15 minutes, a clear straw colored serum sample aspirated by automatic pipette, transferred into clean, dry, labeled tubes, and kept in freezer -20 C: Brain samples were immersed in saline and stored in clean, dry and sterile container until tissue homogenization was conducted to determine the following parameters I) Malondialdehyde (MDA) level II) Glutathione reduced (GSH) level III) Superoxide dismutase (SOD) activity IV) Glutathione peroxidase (GPx) activity V) Catalase (CAT) activity Lipid profile Serum total cholesterol level. Serum triacylglycerol (TAG) level High-density lipoprotein (HDL-C) level Low-density lipoprotein (LDL-C) level Very low-density lipoprotein(vLDL-C) level Apoptosis markers I) caspase 3 level II) BCL2 level Histopathological finding Brains were fully immersed in 20% formalin and stored in clean containers for histo-pathological examination Conclusion The results of the current study revealed that ALA could provide significant benefits from oxidative stress against aging. We found LA supplementation is effective in lowering triglycerides, VLDL, LDL serum levels but not serum cholestrole, HDL.In young rats, LA may produce up-regulation of pro-survival genes. For aged rats, LA may protect the brain cells from apoptosis It can be concluded that ALA can be used efficiently in aged rats.We recomend alpha lipoic acid at dose of 20 mg/ kg diet with aging.
