Abstract

To evaluate the effect of polycaprolactone-based nanofibrous composite scaffolds on adipose derived mesenchymal stem cells. Methods: Adipose derived mesenchymal stem cells (ADMSCs) were seeded onto four groups of electrospunnanofibrous scaffolds with a concentration 3 × 103 cells in a 96-well plate in triplicate. group Ipolycaprolactone (PCL), group IIpolycaprolactone + chitosan (PCL+ CS) , group IIIpolycaprolactone + chitosan + tripolyphosphate (PCL+CS+TPP) and group IVpolycaprolactone + chitosan + tripolyphosphate+ nanohydroxyapatiete( PCL+ CS+ TPP+ nHAp). At first the nanofibers were cut into (10 × 10 mm2) specimens and sterilized under UV light for 3 h. Cells without nanofibers underneath were used as control. The cells were incubated on the scaffolds at37◦C for 2, 5, 7day. Evaluation ofthe cell viability on the prepared nanofibrous scaffolds was done by MTT test,then the results were subjected to one- way ANOVA statistical analysis. The attachment of stem cells to the scaffolds was evaluated by scanning electron microscope. Results: Control group showed that the viability of the adipose stem cells alone without any type of scaffolds was less than that with all the scaffold types at all the time points and it also decreased by time, while the viability and attachment of the other groups increased at 2, 5 days then they began to decrease at 7 days except in group II where it continued in increase after 7 days. group IVwas considered the highest value at 5 days. Conclusion:Polycaprolactone with addition of chitosan and nanohydroxyapatiete could be considered a good scaffold for support of the adipose derived mesenchymal stem cells.
