Abstract

English Summary
Physical studies and photoanalytical techniques for assessment of
some important compounds
The study presented in this thesis includes development and the combination between some
techniques and technologies such as Analytical chemistry, Biochemistry, Photochemistry and
Nanotechnology for the determination of some important compounds such as Alpha amylase
Enzyme, Cortisol Hormone and Nalbuphine Drug in the human serum.
The thesis is divided into four chapters:
Chapter One:-
Includes a general introduction on lanthanides, optical sensors and other methods used for
determination of Alpha amylase Enzyme, Cortisol Hormone and Nalpuphin in the human
serum .
Chapter Two:-
Describes a new nano-optical sensor Tb (III) - carboxyphenyl-18-Crown-6 complex doped
in sol-gel matrix for assessment of Alpha amylase activity in human serum and using the
alpha amylase as biomarker for the early diagnosis of pancreatitis & parotitis for human. The
optimal experimental conditions such as solvent, pH and molar ratio corresponding to
maximum luminescence intensity were investigated. Under optimal conditions, the
luminescence intensity of Tb3+- carboxyphenyl-18-crown-6 doped in sol gel matrix is
inversely proportional to the concentration of 2-CNP in range ( 3.0 X10-4 – 8.0 X 10-9) mol
L−1, ( r = 0.999 ). The detection limit(LOD) (S:N=3) and quantitation limit of detection
(LOQ) are (1.2 x 10-9 and 3.6 x 10-9) mol L-1 respectively. This method is used for the
determination of alpha amylase in human serum.
Chapter Three:-
Describes novel, simple, sensitive and selective spectrofluorimetric method for the
determination of Cortisol hormone serum samples using optical sensor Quinoldine
derivative in acetonitrile. Cortisol can remarkably quench the luminescence intensity of
Quinoldine derivative in acetonitrile at pH (5.7) and λex= 340 nm. The quenching of
luminescence intensity peak of Quinoldine at λem = 617 nm is proportional to the
concentration of Cortisol. The dynamic ranges found for the determination of Cortisol
concentration are (1.0×10-9– 1.0×10-4) mol L-1. The limit of detection (LOD) and
quantification limit of detection (LOQ) are (3.4 x 10-10and 1.0 x 10-9) mol L-1 respectively.
Chapter Four:-
The efficiency of excited-state interaction between Tb3+and the 4’carboxybenzo-
18crown-6-ether (CCE) has been studied in different solvents and pH. The high
luminescence intensity of Tb-complex in DMF at 545 nm was obtained. The photo physical
properties of the green emissive Tb3+complex have been elucidated. The Tb- CCE was used
as photo probe for the assessment of Nalbuphin HCl in the pharmaceutical formulations and
serum samples at pH 6.5 and λex = 285 nm with a linear range 5x10-8 to 1.2x10-6 mol L−1 of
Nalbuphin HCl, correlation coefficient of 0.993 and detection limit of 9.4 x10-9 mol L−1
