Abstract

This research was conducted to fulfill the following objectives; 1) Isolation and identification of some Trichoderma isolates on morphological and molecular basis from Menoufia governorate, Egypt, 2) Evaluation and selection of the best isolates as biocontrol agent against some phytopathogenic fungi, 3) Protoplast isolation from the selected isolates and carry out protoplast fusion among them and 4) Investigation the possible enhancement of obtained fused genomes as biocontrol agent on some phytopathogenic fungi in vitro and in vivo. To fulfill these objectives twenty five Trichoderma isolates were obtained from 105 soil sample from nine districts in Menoufia governorate , Egypt .They are two isolates from Al-Sadat city, one isolate from Ashmoun , three isolates from Elbagour ,one isolate from Barket El sabee, eight isolates from Tala , one isolate from Shebeen El Kom and eight isolates from Menuf .Those isolates were identified on morphological and molecular characteristics to select and improve them to be used as biocontrol agent against some phytopathogenic fungi namely; Alternaria alternata, Fusarium oxysporum, Rhizoctonia solani and Sclerotium spp .
The morphological characters that were useful in the species identification were microscopic and macroscopic. Macroscopic features included growth rate, colony color, colony reverse color, colony edge, mycelial Form and color. The growth rate at various degrees of temperature i.e. 20, 25 ,30 and 35 ℃ on PDA and SNA media, the size of colony are measuring. Microscopic features were conidiophores, conidia, chlamydospores, phialides and branching .
