Abstract

Background. Obesity is a worldwide medical problem, which is an abnormal or extensive fat accumulation. Nowadays, there are many types of nonprescribed dietary supplements that are used for weight loss as Green coffee bean extract and Agiolax. As a result of their effect on fluid excretion, both of these products may be expected to have a damaging effect on parotid salivary gland.
Objectives. The present work aims to compare between the effect of using Green coffee bean extract and Agiolax on the integrity of parotid salivary glands of albino rats.
Material and methods. Twenty-seven adult male albino rats were randomly assigned to 3 groups. Nine received distilled water daily for 2 months by oral gavage (the control group); the other 2 groups received 1 mg/100 gm.body weight green coffee bean extract or Agiolax 8 mg/100 gm.body weight daily for 2 months by oral gavage (the GC and Ag groups, respectively). The parotid glands were dissected and examined histologically, histomorphometrically, biochemically, immunohistochemically for caspase-3 and by real time PCR.
Results.The immunoreaction of caspase-3 was significantly increase in agiolax group and green coffe bean extract. Biochemically, the value of calcium in serum was the lowest in Agiolax group. However, the highest mean value of calcium in urine was recorded in Agiolax group as compared to the other two groups. In addition, the calcium level in tissue was decrease in Green coffee group and agiolax. PCR for Calbindin-2 recorded the lowest value in Green coffee bean extract group.
Conclusions. Long-term oral administration of green coffee bean extract and Agiolax might lead to degenerative changes in the parotid glands. A greater deleterious effect was caused by agiolax, as it caused more caspase-3 expression, significantly reduced Ca level in the tissue and in serum. In addition, Calbindin-2 recorded the lowest value in Green coffee bean extract group.
