Abstract

Summary
This study aimed to assess seminal Sirt-1 expression in infertile men before and after varicocele repair. Overall, 53 men presented to the University hospitals after ethical approval and informed consent were investigated.
They were allocated into:
I- Fertile healthy men (n=23) as controls
2-Infertile OAT men associated with varicocele (n=30)
Inclusion criteria: Stable relationship, confirmed abnormal semen parameters and normal female partner.
Exclusion criteria: Leucocytospermia, hormonal therapy, immunological disorders, dyslipidemia, hypogonadism, cardiovascular disorders, morbid obesity, and hepatic/renal failures.
They were subjected to:
History taking
Clinical examination
Semen analysis according to WHO guidelines (2010).
Estimation of seminal SIRT-1expression by Western Blot technique.
Surgical repair of varicocele followed up for 3 months.
The results were as follows:
Seminal SIRT-1 showed significant decreases in infertile OAT with varicocele compared with fertile men. Also, seminal SIRT-1 showed significant increases in infertile OAT with varicocele 3 months post-operatively compared with the pre-operative data.
Seminal SIRT-1 (ng/ml) showed significant decreases in infertile OAT men with grade III varicocele compared with infertile OAT men with varicocele grade II.
Seminal SIRT-1 showed significant increases in infertile OAT men with varicocele post-operatively compared with the pre-operative data
Seminal SIRT-1 expression showed significant positive correlation with sperm concentration, sperm motility, sperm normal forms and nonsignificant correlation with the age.
Conclusion
Seminal Sirt-1expression has a role of male infertility being significantly decreased in infertile OAT men associated with varicocele and significantly increased after varicocele repair.
