Abstract

In the present study, we have evaluated the cell viability of human HCT-116 cell line and healthy cells by MTT. Determination the levels of expression of (P53, BAX, BCL-2 & Caspase-9) in HCT -116 cell line, Using Real Time – PCR .
In laminar flow safety cabinet the human HCT-116 colon cell line was cultured in DMEM containing heat-inactivated fetal bovine serum, streptomycin , penicillin and streptomycin in a humidified atmosphere of 95% air and 5% CO2 at 37 ºC incubator. Then the cells treated with different concentration of nanocurcumin to evaluate cytotoxic effect of nanocurcumin on the HCT-116 cell line and healthy cells by MTT assay and also gene expression of P53, BAX, BCL-2 & Caspase-9 was determined in HCT-116 cell line.
After statistical analysis our results referred to the following:
• Cytotoxic effect of Nanocurcumin on HCT-116 cell line and normal retina cells RPE1:
Our results referred to nanocurcumin inhibited the growth of HCT-116 cells in a concentration-dependent manner. And had no inhibitory effect on normal cells after 48 hrs treatment with different concentrations of nanocurcumin ranged between 0.78 to 100 µmol /ml.
•Analysis of PCR product of P53 gene in HCT-116 cell line by using Real Time – PCR:
The HCT-116 cell lines showed increase in the level of expression of P53 by increasing the concentration of nanocurcumin.
