Abstract

The study was conducted to assess the potential protective effects of curcumin against colistin
induced nephrotoxicity and neurotoxicity. The results were summarized as follow:
I- Effect of curcumin and/ or colistin on oxidant/ antioxidant status of kidney and brain tissues: Intraperitoneal administration of colistin at a dose (300.000 IU/kg b.wt) once daily for 6 successive days induced a significant decrease in renal and brain CAT, GSH levels as well as significant elevation of MDA concentration compared with the control group. While, administration of curcumin concurrently with colistin resulted in a significant increase in renal CAT and brain GSH levels and significant decrease of MDA concentration compared with those of colistin group.
II- Effect of curcumin and / or colistin on inflammatory markers of renal and brain tissue: Effect on TNF-α, IL-6 and NO: Intraperitoneal administration of colistin at dose (300.000 IU/kg b.wt )once daily for 6 successive days showed a significant increase in renal and brain TNF-α, IL-6 and NO concentration when compared with those of control group. Co-administration of curcumin (200mg/kg b.wt) once daily for 6succssive with colistin induced a significant reduction in renal and brain TNF-α, IL-6 and NO concentration when compared with those of colistin group.
III- Effect of curcumin and / or colistin on kidney injury parameters: Colistin administration a dose at dose (300.000IU/kg b.wt )once daily for 6 successive days induced a significant elevation in serum urea, creatinine and uric acid compared with those of control group.
IV- Effect on brain GABA concentration: Interperitoneal administration of colistin at dose( 300.000IU/kg b.wt )once daily for 6 successive days showed a significant increase in GABA concentration compared with those of control group.
V- Histopathological and immunohistochemichal finding: kidney: Histological findings revealed that in colistin group degenerative changes in renal tubules with tubular dilatation as well as presence of cast in the lumen of some renal tubules were noticed. In concurrent group, the renal tubules showed regenerative attempt. Immunohistochemically, the study observed that in curcumin group the renal tubules gave strong positive reaction with anti-Bcl-2 antibody. In curcumin group, mild expression of caspase was observed in the renal tubules). In concurrent group, no reaction was observed in the renal cortex. While moderate reaction was observed in renal medulla. Brain: In colistin group, degenerated neurons surrounded by empty spaces. In concurrent group, the neurons were nearly similar to control group. In the cerebellar cortex of colistin group the effect were concentrated on the Purkinje cell layer. The Purkinje cells were disappeared in several areas, its normal pyriform shape was lost with deep homogenous cytoplasm.
