Abstract

The present study was conducted during 2015 and 2016 at Genetic and Genetic Engineering Laboratory located in the Branch of Inland waters and aquaculture at Al-Kanater Al-Khyaria, Kalyubeia Governorate, Egypt.
About 100 specimens of the wild Tilapia spp. O. niloticus and C. zillii were collected from different sites of different degrees of salinities in the lakes of Manzala, El-Bardweel and Nasser Lake in December 2015 and January 2016. Fin clips has been obtained and preserved in absolute ethanol then transferred to Genetic and Genetic Engineering Laboratory located in the Branch of Inland waters and aquaculture at Al-Kanater Al-Khyaria. All samples will be kept at -20 ºC until DNA extraction.
For measurement of some of physical and chemical parameters which indicate environmental conditions, Water samples has been collected using 1.5 L Ruttner sampler from 5 sites from each lake. Water Salinity, Hydrological parameters such as temperature (T), dissolved oxygen (DO) and pH value have been measured directly in the field.
For histological study, 5 fish were taken from different salinity degree sites, Fish were dissected, the abdominal cavity.
