Abstract

Paclitaxel is one of the most effective and commonly used anti-neoplastic drugs with activity against several tumors including ovarian cancer, metastatic breast cancer, bladder, testicular, lung, head and neck cancers.Omega 3 polyunsaturated fatty acids are essential for lipid derived modulators, gene expression and inflammatory process as they are not synthesized in the human body, they need to be supplemented through the diet. They have many beneficial effects in some psychiatric and neurodegenerative diseases. Moreover, they attenuate production of proinflammatory cytokines which induce neuropathy. Yellow phenolic curcuminoid have a wide range of biological activities including antihyperglycemic, antioxidant, antihypolipidemic, neuroprotective, anti-inflammatory and antiaging effects.The aim of the current study was to compare between the possible protective effect of omega 3, curcumin and their combined use on paclitaxel experimentally induced peripheral neuropathy in sciatic nerve of adult male albino rats using histological and immunohistochemical study.The present study was carried out on 50 adult male albino rats, their weight ranged between 240 and 260 gm.Animals were randomly divided into 5 groups (10- rats each):
group I (Control group): Rats were injected with saline (1ml) intraperitoneally every other day for 4 weeks.
group II: Rats were treated with paclitaxel (2mg/kg intraperitoneally every other day for 4 weeks).
group III: Rats were treated with paclitaxel (2mg/kg intraperitoneally every other day for 4 weeks and omega 3 (0.5 g/kg daily orally via gastric tube for 4 weeks) starting from the first day of paclitaxel administration.
group IV: Rats were treated with paclitaxel (2mg/kg intraperitoneally every other day for 4 weeks) and Curcumin (200 mg/kg orally via gastric tube for 4 weeks) starting from the first day of paclitaxel administration.
group V: Rats were treated with paclitaxel (2mg/kg i.p every other day for 4 weeks, omega 3 (0.5 g/kg daily orally via gastric tube for 4 weeks) and Curcumin (200 mg/kg orally via gastric tube for 4 weeks) starting from the first day of paclitaxel administration.
Animals were sacrified after 24 hours from the last dose of paclitaxel administration. Sciatic nerve samples were taken from rats of all groups.
Tissue sections were prepared for light microscopic examination by staining with Hematoxyline &Eosin, osmic acid and immunohistochemical staining for neurofilament. Also ,sections were prepared for electron microscopic examination.
