Abstract

Eight different infectious coryza vaccines were prepared in this study. Four vaccines formula with different adjuvants (liquid paraffin oil, MontanideTM ISA 71, MontanideTM ISA 201 and MontanideTM Gel01) and four vaccines formula by using Date palm (Phoenix dactylifera L) and Mentha piperita immunomodulators included in liquid paraffin oil adjuvant infectious coryza vaccine only in ratio of 1mg or 2mg of each immunomodulators, these 8 vaccines were used in vaccination of 8- 10 weeks old chickens and evaluated by using Microplate agglutination test (MAT) and ELISA. Results revealed that the all prepared vaccines were sterile and safe. The humoral immune response of chickens against the four vaccines formula yielded that, vaccine no. 1, 2, 3 and 4 prepared with different adjuvants developed agglutination antibody titers against each of the three tested strain antigens of Av. paragallinarum beginning from the 2nd week post- vaccination, reaching peak at 14- 16th week, 10- 12th week for vaccine no. 1 and 2 respectively and 6- 8th for vaccine no.3 and 4 respectively. It was cleared that, MontanideTM ISA 71VG oil adjuvant infectious coryza vaccine yielded maximum rate of antibody titer higher and faster than liquid paraffin oil adjuvant infectious coryza vaccine, MontanideTM ISA 201VG oil adjuvant infectious coryza vaccine resulted in peak of antibody more higher and faster than MontanideTM ISA 71VG oil adjuvant and liquid paraffin oil adjuvanted infectious coryza vaccine respectively, in addition to, MontanideTM Gel01 adjuvant infectious coryza vaccine yielded more faster and earlier antibody peak than vaccine no. 3, 2 and 1 respectively, as well as higher than vaccine no. 2 and 1 respectively. The results of ELISA for evaluation of humoral immune response of chicken against vaccine no. 1, 2, 3 and 4 were generally came parallel with results of MAT . The challenge test results revealed that, immunized chickens were significantly protected in relative to unimmunized control chickens supporting the results of MAT and ELISA. Evaluation of the humoral immune response of chicken vaccinated with vaccine no. (5, 6, 7 and 8) by using MAT, ELISA , cleared that agglutination antibody titer against each of the three tested strain antigens of Av. paragallinarum beginning from 2nd weeks post immunization continued to increase at relative rates against the three antigens (W, Modesto, 0222) reaching maximum rates at the 10th to 12th week post immunization then gradually declined to reach their lowest levels by the 24th week post vaccination, the data also cleared that using of 2mg of either Date palm or Mentha piperita included in the vaccine yielded levels of antibody titers higher than using 1mg of either Date palm or Mentha piperita. The results obtained in this study concluded that, the infectious coryza vaccine adjuvanted with either MontanideTM ISA71VG, MontanideTM ISA 201VG or MontanideTM Gel 01 adjuvant yielded immune response in chicken better than liquid paraffin oil .Using Date palm and Mentha piperita as immunomodulators included in liquid paraffin oil adjuvant infectious coryza vaccine can enhance immune response of chicken against infectious coryza disease .
