Abstract

The present study was conducted to evaluate the protective effect of four different nephroprotective agents, two of them were synthetic compounds (TSA and MS-275), and the other two were of natural origin (Fenugreek and bee honey) against cisplatin nephrotoxicity. This experiment was performed using 260, 8-10 week old male C57BL/6 mice. Mice were divided into equal groups each of ten. group I control, group II cisplatin, group III Cisplatin+ TSA, Cisplatin+MS-275, Cisplatin+Fenugreek and Cisplatin+bee honey, group IV TSA, MS-275, Fenugreek and bee honey. Animals were anesthetized and sacrificed 72hr after cisplatin injection. Blood and kidney tissues were collected. Kidney tissues were processed for histology, TUNEL assay, neutrophil, protein and RNA isolation. Our result showed that TSA, MS-275, Fenugreek and bee honey were able to suppress cisplatin induced kidney dysfunction as seen by decrease serum creatinine which associated with less tubular necrosis, cast formation, preservation of brush border and less apoptosis as compared to cisplatin treated group. They also showed a significant decrease of kidney inflammatory markers including cisplatin induced neutrophil infiltration and cisplatin induced increase in TNF-, IL-6 and MCP-1 in kidney, plasma and urine as compared to group II (cisplatin).
