Abstract

Two experiments were carried out during the period from 2013 to 2017. The first experiment was achieved in the tissue culture and Biotechnology laboratory and green house with project of Improvement of the Main Vegetable Crops and Hybrids Production (IMVCHP), Horticulture Research Institute, Dokky, Giza, Egypt to determine the appropriate growth regulator concentrations and different combinations to establish protocol of callus induction and its regeneration ability using different vegetative organs, i.e. runner, leaf, anther culture and immature fruit from three strawberry cultivars, i.e., Festival, Fortuna and Sweet charlie. The second experiment was carried out at a private farm in Badr district, El- Behera governorate for evaluation of field performance of strawberry plants obtained by callus induction using different explant types and also RAPD markers are used for identifying of different strawberry new strains.1- In view of the obtained results, it was found that using the runner as explant source for both Festival and Fortuna cultivars and Murashige and Skoog media supplemented with 0.5 mg L-1 BAP + 1.5 mg L-1 2,4- D was very efficient in inducing callus production with the highest percentage. 2- Also, MSR8 (2.0 mg L-1 BAP) was the best media for shoot regeneration percentage (%) and shoot length. For rooting, half MS supplemented with 1.0 mg L-1 IBA resulted the highest roots percentage, roots number and root length. 3- The obtained results indicated that RAPD markers were an efficient for varietal identification and classification in strawberry. The polymorphism resulting from RAPD markers among the parents and the resulted selected strains confirmed that A1 and A9 strains resulted from Festival cultivar were superior in respect of growth related parameters and yield components.
