Abstract Aerosol therapy is the adminstration of drugs in the form of inhaled fine particles. When a very low percent, only about (1-10%) of nebulized medication reaches the lung of the patient after end of drug nebulization, this results in less effective treatment and also higher cost for very expensive drugs The main aim of this study was to determine fill volume change effect on salbutamol delivery to patients on single limb NIV using two different aerosol producing devices (Jet and Vibrating mesh nebulizer). Another aim Alsowas to determine the effect of nebulizer design (Six different designs were used) on drug delivery, as a step for optimizing aerosol delivery to the lungs of mechanically ventilated patients and establishing of practical guidelines for the optimum fill volume of inhaled solutions. Results of study indicated that, JN design had a great effect on emitted dose while different designs of VMN had no significant effect. At the same fill volume VMN resulted in better lung deposition and systemic absorption at shorter nebulization time than JN indicating high efficacy of VMN. Increasing fill volume is an essential factor for improving aerosol delivery by JN due to its significant effect on lung deposition and systemic absorption. Also amount of drug measured in inhalation filter increased significantly by increasing fill volume in both In-vitro and Ex-vivo study. VMN is not significantly affected by increasing fill volume due to lower residual volume, and still has a better aerosol delivery than JN using any fill volume. So it is recommended to increase fill volume with JN (4ml) but not recommended with VMN to be increased than (2ml). |
