Abstract This study was conducted on Periwinkle plant as a medicinal plant that aimed to development of tissue culture efficient protocol to get the highest alkaloid content and vincristine accumulation that used in the treatment of cancer. Five different ecotypes of Egyptian Catharanthus roseus were selected, namely (Ismailia, Fayoum, Alexandria, Sharkia and Cairo) for callus induction, alkaloids production and vincristine accumulation. The results showed that Ismailia and Fayoum ecotypes were the best of all ecotypes in all studied morphological characters and total alkaloids content (TAC). The results of callus culture showed that M1 medium (2, 4-D) 1.0 μM and (BA) 0.5 μM) gave the highest value for callus induction frequencies and callus fresh weight for produced callus from leaf and stem. The results also showed that Fayoum ecotype was the highest content of alkaloids (2.22 mg / g) for produced callus from stem explant which gave on HPLC unit the highest content of vincristine 3.83363 µg/g, which represents 10.84 fold compared to the original plant. The results confirm superiority of the Fayoum ecotype in alkaloids production and vincristine accumulation than all studied ecotypes. |
