Abstract

In the present study, the sealing ability of the three obturation systems (RealSeal SE, Activ GP and AH Plus sealer with gutta-percha) was measured after final rinsing with EDTA, Biopure MTAD and QMix 2in1. Micro-leakage test was performed by bacterial leakage method using Enterococcus faecalis as a bacterial marker.
One hundred extracted human maxillary single rooted teeth were collected for this study. Ten prepared root canals were randomly selected for the two control groups (n=5) and the remaining ninety root canal were randomly divided into three groups (n=30) according to the final rinse used for each group (17% EDTA, Biopure MTAD, QMix 2in1). Each group was later subdivided according to the obturating system used into three sub-groups (n=10) (RealSeal SE, Activ GP, AH Plus/gutta-percha).
Obturation with different materials was performed following irrigation with the tested irrigating solutions. The roots were stored for one month, and then suspension of E. faecalis was applied into the coronal portion of the roots and incubated within Eppendorf tube containing trypticase soy broth (TSB) for one week, one month and three months testing periods. After incubation, cultures from each suspension broth were obtained to estimate colonies of bacteria that leaked from the apical portion of the roots obturated with different materials.

The result showed that, till one month, the RealSeal SE was significantly better than Activ GP and AH Plus in sealing of the root canals. However, at three months, no significant differences were found, in the sealing ability, between the three obturating systems. Biopure MTAD significantly enhanced the sealing ability of the three obturation systems compared to 17%EDTA and QMix 2in 1.
Within the parameters of this study the following conclusions could be drawn:
1) Real Seal SE obturation system appeared to offer initial better sealing ability, of the root canal system, compared to Activ GP and AH Plus /gutta-percha obturation systems. However, by time, the three obturating systems were comparable in the sealing ability.
2) Irrigation with Biopure MTAD may enhance the sealing ability of obturating systems than both 17% EDTA and QMix 2in 1.
Recommendation:
The new monoblock obturation systems (RealSeal SE, Activ GP) are not found to have better sealing ability than conventional obturating systems(AH Plus/gutta-percha)
