Abstract

In the present study, experiments were carried out to search for new natural antibiotics through isolation of various Actinomycete strains from different soil samples from Bayt al-Faqih area - Al Hudaydah governorate- west of Yemen and studying the antimicrobial effects of metabolites they produce. The isolate (S37) which was found to produce a wide spectrum antimicrobial agent was selected and identified. The cultural, physiological and biochemical characteristics of the isolate identified the strain as a member of the genus Streptomyces.
The parameters controlling the biosynthetic process of the antimicrobial agent including; different pH values, different incubation period, and different carbon and nitrogen sources, were fully investigated and the antibiotic was produced at the optimum condition. The antibiotic was extracted using chloroform: ethyl acetate (1:1). The purification of the antibiotic was performed using column chromatography technique. The purified antibiotic produced by isolate S37 was given the name Faqihmycin.
