Abstract The aim of the study was to compare the cytotoxicity and the flow properties of Triple antibiotic paste versus propolis as an intracanal medicament. Pastes of Triple Antibiotic pastes were prepared and and sterilised by ethylene oxide gas and soaked in E-MEM for 7 days .Samples were placed in a dark container at 37 °C and 100%. Cytotoxicity was evaluated using the MTT assay at 24 , 48 and 72 hours, IC50 values were calculated as well as the therapeutic dose cytotoxicity values. Flow properties were evaluated by putting the pastes between two parallel glass slabs and inserting a weight of 500grams on the parallel slabs , diameter of the circles formed were measured and calculated using a vernier ruler. Data was calculated and statistically analysed. MTT results showed statistically insignificant difference between Triple antibiotic paste when evaluated at 11 different doses .When IC50 values were calculated from readings obtained from MTT assay results showed statistical significant differences with the lowest IC50 value recorded was for Propolis at 48 hours. When cytotoxic action of therapeutic values was tested from MTT assay readings , results showed significant statistical differences (P value : 0.0008) between both propolis and Triple Antibiotic Paste with highest viable cell percentage to triple antibiotic paste with 87.53% vitality at 48 hours followed by 76.63% vitality at 72 hours of TAP followed by 76.32 vitality of TAP at 24 hours then Propolis with a low cell vitality of 34.21% at 72 hours then 32.54% vitality at 48 hours and the lowest cell vitality percentage was found for Propolis at 24 hours 20.5% suggesting it to have the most cytotoxic action. Flow rate results showed insignificant statistical difference between flow of Triple antibiotic paste and Propolis paste. |
