Abstract

Umbilical cord blood are cheap,easily,and rich source of hematopoietic stem cells.
Umbilical cord blood has been widely used as a source of hematopoietic stem cells to treat a variety of genetic, hematologic, immunologic, metabolic, and oncologic disorders.
Recovery rate of CD34+ cells should therefore be monitored to ensure that significant cells will not be lost during separation of cord blood. This will improve the qualification for a transplant and success rate from the cord blood bank‘s point of view.
The present study aimed to evaluate different obstetrics circumstances and laboratory factors affecting umbilical cord stem cell collection and cryopreservation as nucleus for clinical application: ”phase I study.40 UCB units were collected from 40 healthy females after their consent and complete lab.investigation .Blood volume was measured and MNCs were separated using Ficoll hypaqe. After that, CD34+ cells were separated by magnetic field Separation.
Both of MNCs and CD34+ viability tested, counted and cryopreserved using cryoprotecting medium at -196oC for 1 year through which one sample was thawed after one month, three months, six months, 9 months and 12 months respectively and viability test was done.
Data of the present Study showed that percentage of cell viability after thawing not significant affected throughout cryopreservation period, this means that components of cryoprotectant medium was effective in protecting cell viability throughout cryopreservation period.Quality of only when cells are not significantly lost during processing.
